Just follow these simple steps.
1. Either install any DO FOLLOW Plugin in your wordpress. which you can get just by Goooogling.
2. Or if you want to do manual tweaks in your files to achieve this then open the file “wp-includes/comment-template.php”
Now
(For WordPress 2.3 or older: line 48)
(For WordPress 2.5: line 114).
(For WordPress 2.6: line 116)
REPLACE :
$return = "<a href='$url' rel='external nofollow'>$author</a>";
with this:
$return = "<a href='$url' rel='external'>$author</a>";
Please observe this image carefully.
NOTE : Remember to take regular backups of your all files from server before doing any changes to files, i am not responsible for any side effect. 
Now Done, You have hacked your wordpress to have DO Follow comment links !!
I recommend all blogger to use this tweak to give proper rewards to the people who comment on our blog !
* UPDATED : Currently my blog comments are NO FOLLOW, i actually suffer a lot from the comment spamming on my blog, so i decided to remove the DO FOLLOW from comment and makes to back to No Follow, your can get more information about comment spamming from here and here. actually i read those two posts and decided this . sorry guys.

you can install akismet to take over the manual method of checking for comments. also, it is a good thing that comments appear instantly. some folks don’t like the idea of waiting for their comments to be moderated.

The replacement line you have posted above contains special characters. One of my client took your line and pasted in line 116 and his blog was screwed up. See what happens when I paste the live below:
$return = “$author”;
The double quotes and single quotes are all special characters and fail in PHP parsing. You should put the original line and modified line in plain text code so that people don’t get messed up.

Ohh,I really sorry if your client face any serious problem due to this. now i corrected the error, and added the Image to justify what actually u have to do to make them Do Follow. Actually i allready given both the actual code and modified code there.
Please have a look , and suggest if its still wrong.

I have got also some blogs,most people send spam comments.
Always carefully control commenters and do not apply spam commenters or only writing short words like thanks etc. I read your background,you are computer postgraduate student and I believe that you will be a big Specialist in your field.The Indians are also very good in Computer,Information Systems Area.

Thanks for the info. I am going to find a plugin to use. How in the world can I find out if someone’s blog I am commenting on is indeed a do-follow blog? I am working hard on building links and it is taking forever.

The NoNoFollow or U Commnet I Follow movement is a great way to increase blog participation at the end of the day – if you want to leave a comment the last thing you want is for it to be moderated by some backdoor piece of code. I for one have implemented the DoFollow plugin on my blogs and hope more people do so in the future.
